枯草菌蛋白質分泌系で作用するSRP様粒子のシャペロンとしての機能解析

研究概要	枯草菌におけるα-アミラーゼ、プロテアーゼ、セルラーゼなどの有用酵素を大量に分泌する経路でSRP様粒子がシャペロンとして働いていることを分子生物学的に明らかにするために、SRP様粒子の構造と機能を解析した。これまでSRPはヒトやイヌの細胞で主に研究されてきた。ヒトSRPは1分子のRNA(SRP 7S RNA)と6種類の蛋白質から構成される。枯草菌におけるsmall cyto plsmic RNA(scRNA、271塩基)はSRP 7S RNA(300塩基)の相同物質であり、二次構造上類似している。一方大腸菌においてSRP 7S RNAの相同物質として4.5S RNA(114塩基)が検出されているが、分子量は約1/3であり、SRP7S RNAの一部分と構造上類似している。枯草菌とscRNAはドメインI、II、IVより構成され、ヒトSRP 7S RNA(ドメインI〜IVで構成される)に比べてドメインIIIが欠損している。ドメインIVは全ての生物に共通しており、機能的に必須のものを予想される。一方、ドメインIはヒト、枯草菌には共通であるが大腸菌4.5S RNAには含まれず、その機能が注目される。枯草菌scRNAのうち、ドメインIを欠損させると耐性胞子が形成されなくなる。そこでscRNAドメインIに結合する蛋白質を枯草菌細胞抽出液から純化したところ、既知蛋白質HBsuであった。HBsuは本来はDNA結合蛋白質であるが、二本鎖RNAにも結合できることを明らかにした。またHBsuの構造はヒトSRP 7S RNAのドメインIに結合するSRP9/14複合体の構造と類似していることを明らかにした。
