複数のRNA結合タンパク質の包括的解析とホヤ卵内母性mRNAの高次機能の解明

研究概要

ホヤ卵内には、あらかじめ局在する分化決定因子の存在が知られている。そのホヤ卵内で発現しているRNA結合タンパクを解析し、母性mRNAとその翻訳制御が初期発生に果たす役割を明らかにする事を目的に、これまでに以下α種類のRNA結合タンパク質遺伝子を単離し、その発現様式を明らかにした。
1 :CiYBl,2,3:よく保存されたcold shock domainを持っており、マスキングタンパクとして知られるY-bOXproteinのホモログである。3つのアイソフォームは1つの遺伝子から選択的スプライシングにより作られる。ゲルシフトアッセイにより、in vitroで、Gに富むRNAへの結合能があることを示した。
2 :CiHul;RNA recognition motif(RRM)型のRNA結合ドメインを3つ持ち、神経分化に関わるHuDと比較的高いアミノ酸配列の同一性を示した。卵内で高い発現を示し、尾芽胚期においては、神経細胞では発現せず、間充識細胞でのみ強い発現を示した。
3 :CiMsil;MusashiタイプのRNA結合タンパク質である。N末側にRRMを2つ持つ。卵内でも高い発現を示し、のう胚期以降予定神経細胞でzygoticな発現が見られはじめ、神経管形成時の神経細胞で高い発現を示す。
4 :CiRGG1;RGG box(Arginine-Glycine-Glycineの繰り返し配列)とRRMを持つ。未受精卵で高い発現を示し、2細胞期のごく短期間胚の前側へ局在する。このような局在性を示す分子は、これまで知られていない。
5 :CiGRPl,2;コールド・ショックにより誘導されるGlycine-rich RNA結合タンパク質のホモログと考えられる。未受精卵および初期卵割期と神経胚期以降の脳で高い発現を示した。
