急速凍結レプリカによる個々のミオシンクロスブリッジ構造変化の高分解能3次元像解析

研究概要	筆者は、高い空間/時間分解能を有するマイカ細片急速凍結フリーズレプリカ電子顕微鏡法を駆使して、アクチンの滑り運動を起こしているミオシン・クロスブリッジの一瞬の姿を捉え、筋収縮や種々の細胞運動の源となる滑り運動の分子メカニズムを構造の面から追求してきた。既に、マイカ表面に吸着したアクチン・重メロミオシン硬直複合体にATPを添加する前後に急速凍結して得られた像を比較し、クロスブリッジのコンフォメーションに大きな変化があることを示してきた。本年度は、滑り運動中のクロスブリッジの更に詳細な3次元構造を追求することを目標とした。最終的には、レプリカ試料を広範囲に傾斜して像を撮影して3次元像再構成することを目指しているが、その途中過程において、一連の像を用いてアニメーションを作成したところ、それだけでも3次元構造が良く認識できるばかりか、X-線結晶モデルで見られるサブドメインの配置さえ明瞭に認識できることが判明した。硬直状態においては、ミオシン頭部の上下50KD-ドメインはアクチンフィラメントにほぼ直角に配位し、それぞれが隣接する2個のアクチンに結合すると考えられている。滑り運動中のクロスブリッジの像(主としていわゆる"weakly-bound state")を実際に観察すると、それらはむしろ平行に配位していた。さらに上位50KD-ドメインの先端の小さなドメインのみがアクチンと結合し、50KD-ドメイン本体とは細い紐状の構造でつながっていた。少数のクロスブリッジにおいては硬直状態と類似の結合様式も見られたが、尻尾の方向はむしろADP型に近く、真の硬直結合型は今のところ見つかっていない。硬直状態の構造解析が急務である。また滑り運動中には様々のコンフォメーションが混じっているのは自然であり、更に多くの粒子のコンフォメーションを解析する必要がある。観察結果からは、"lever-arm"仮説による滑り運動機構の説明は困難と思われる。
