研究概要

本年度はアルツハイマー病における機能修復実験系の開発のためにβアミロイド蛋白の神経毒性による神経細胞死と機能修復の、より良いモニター系開発を目指した。
まず、神経細胞死の直接の原因を探る、すなわち神経毒性のメカニズムとして、βアミロイドがつくるチャネルが非特異的カチオンチャネルであることから、Ca^<2+>の細胞内流入をモニターした。測定にはCa^<2+>蛍光色素fura2とビデオつき蛍光同時多点解析装置を用いた。βアミロイドは濃度依存的に神経系由来のGT1-7細胞の細胞内Ca濃度を上昇させた。このCa^<2+>上昇はβアミロイド濃度依存的であった。この実験系は一つ一つの細胞によるレスポンスの差も多数の細胞間で比較することができ、メカニズム研究としても優れている。
