研究概要

GPIアンカーの生合成とタンパク質への転移に関与する遺伝子のうちDPM3サブユニットの遺伝子をクローニングし、これがDPM1とDPM2の仲立ちをしている制御サブユニットであることを証明した。固形癌細胞、特に消化器癌では、シアリルルイスaがシアリルルイスxと並ぶ重要なセレクチンのリガンドとして機能し、その発現が癌の血行性転移および患者の予後と深く関連することを明らかにした。白血球系細胞におけるセレクチンのリガンドであるシアリルルイスsの発現が、フコシルトランスフェラーゼVIIによって調節され、細胞の悪性化や刺激に際してこの酵素遺伝子の転写が活性化されることを明らかにした。NeuGcの発現を律速支配している酵素CMP-NeuAc水酸化酵素を精製・クローニングした。またa-1、6-GlcNAc転移酵素遺伝子導入マウスを用いた解析より約100kbpの遺伝子断片が転写を制御していることを明らかにした。GM2/GD2合成酵素遺伝子ノックアウトマウスを用いた解析により、当酵素は神経組織の構築維持と損傷時の修復・再生にとって重要であることが示された。また新規シアル酸転移酵素遺伝子のクローニングと機能解析を行った。GD3合成酵素遺伝子を誘導する系を用いて当酵素の発現によって新たに発現する遺伝子をDfferential display法により探索したところDAP群を発見した。新たにクローニングしたFUT9は既知のal,3フコース転移酵素と比較してルイスx合成活性が強いという特徴をもっていることがわかった。GlcNAc-6-硫酸特異的bl、4GalTを同定、精製した。また大腸粘液癌特異的GcNAc1,6SulTを同定した。硫酸化糖脂質生合成のキー酵素であるCSTノックアウトマウスを作成・解析したところ生後6週より下肢の対麻痺が出現し、精巣も野性型の5分の1程度であることが観察された。
