研究概要

本研究班では4つの異なるが互いに関連したアプローチからエイズの予防・制御を目指した基礎研究を行ってきた。すなわち、1)新規抗ウイルス薬の開発、2)薬剤耐性の機序とその克服、3)免疫療法についての基礎研究、4)免疫不全の進行にかかわる宿主因子である。本年度の成果として馬場らは種々の物質のTatに対する抑制効果を検討し、streptomyces属より単離された新規物質K12にtatを介したHIVの転写を抑制する活性を発見した。満屋らは逆転写酵素阻害剤に対する多剤耐性変異ウイルスのフィットネスについて新しく開発した増殖競合試験を用いて検討し、耐性獲得で失われたフィットネスを回復する変異株が出現することを明らかにした。満屋らはさらに本邦においても逆転写酵素やプロテアーゼに多くの耐性変異が出てきていることを報告した。高松らは治療に用いられる抗ウイルス剤の種類によって薬剤耐性変異株の挙動に変化を認めることを報告した。松下らは多剤併用療法導入後の抗HIV免疫応答を解析し、Viral loadが低下するとむしろ細胞傷害性T細胞活性が低下すること、中和抗体価は上昇する症例があることを示し、多剤併用療法下に抗HIV免疫を強化する治療法を行う意義を明らかにした。一方、宿主因子の研究として志田らは脳内でのウイルスのりザーバーとして働くアストロサイトでのrev機能不全の原因を探ったところ、主要コファクターであるhCRM1が存在せずに、そのVariant(u-CRM1と命名)が発現していることを見出した。
以上の研究によりエイズ制御に向けた病態の理解は深まったと考えられるが、さらに来年度は具体的な制御に向けて踏み込んだ研究を展開する。
