研究概要

(1) 抗マウスFRP-1mAbsの作製とその機能の解析
最初にマウスFRP-1を構成的に発現しているHeLa(HeLa/mFRP-1)細胞を樹立した。L929細胞をラットに接種し、免疫脾臓細胞とマウス形質細胞腫細胞を融合し、HeLa/mFRP-1細胞を抗原にして、抗mFRP-1抗体を分泌しているhybridomaをスクリーニングした。全部で19種の抗mFRP-1モノクローナル抗体を作製した。mFRP-1はalloantigenであり、そのalloantigenicityは1アミノ酸の違いに起因することが明らかになった。HeLa/HIVgp160細胞とL929/hCD4/hfusln細胞との混合培養の系に抗mFRP-1抗体を添加すると、3種の抗体が、多核巨細胞形成を阻害した。
(2) CD98のlight chainの遺伝子クローニング
CD98/4F2のlight chainの遺伝子クローニングを行ったところ、507アミノ酸から構成され、11-12回膜を貫通している蛋白であることが明らかになった。アミノ酸トランスポーター活性を有すると思われる。その遺伝子は第16染色体に位置していることを証明した。
(3) FRP-1 ligandのクローニング
現在までの所、FRP-1 ligandのクローニングには成功していないが、骨肉腫由来細胞にFRP-1 ligandが高率に発現していることを突き止め、遺伝子クローニングを行っている最中である。
