造血システムにおける自己複製と分化機構の解析

研究概要

「造血システム」公開班会議は、4班合同で行い(平成11年11月19日)、一部の成果は、日本血液学会国際シンポジウム(平成11年4月19日)で発表した。この2年間の主たる成果は以下の通りである。
i)胎生期における造血発生を解析し、造血幹細胞は血管内皮細胞との共通祖先細胞から分化し、両者の間に緊密な相互作用があることを明らかにした。この成果は、造血システム研究のみでなく、血管新生の研究にも大きく貢献するものである。
ii)胚性幹細胞(ES細胞)から血液細胞・血管内皮細胞への分化を制御することが可能となった。
iii)分離した造血幹細胞1個を放射線照射したマウスに連続的に移植することにより、その高い自己複製能を示した。
iv)ES細胞の自己複製能と多分化能維持にgp130を介するSTAT3の活性化が必須であること、またSTAT3は、BMP下流の転写因子Smadと協調することを明らかにし、幹細胞の自己複製を分子レベルで解析できるようになりつつある。また、サイトカインのシグナル伝達において負のシグナル伝達機構(CIS/JAB/SOCS/SSI)を、個体レベルで明らかにした。
v)GATA-1,GATA-2など造血制御因子の制御機構をトランスジェニックマウス法を用いて明らかにすることができた。Evi-1転写制御因子が増殖抑制・細胞死抑制に関与することを示し、造血幹細胞の悪性化機構を解明した。
vi)造血幹細胞への遺伝子導入・移植実験をよりヒトに近いサルなどを用いて行うことができるようになり、臨床応用への端緒を開いた。
