研究概要

21世紀の医療として、再生医学が、そしてそれを可能にする「幹細胞」が注目されている。幹細胞は、分化して多種類の細胞をつくる多分化能と同時に、幹細胞自体を生みだす自己複製能を有する。
本研究班では、以下の進展がみられた。
1)第一班を中心に他班からも多数の研究グループが造血発生の研究に参加し、大きな成果をあげた。造血細胞が、血管内皮細胞から発生を共にすることが示され、幹細胞発生を試験管内でアプローチできるようになりつつある。これは、幹細胞が自己複製能を獲得する過程を明らかにするだけでなく、血管新生機構の理解にも大きく貢献する。第二班、第三班のシグナル・転写研究は、着実な研究の進展を示し、サイトカインシグナルを抑制する分子の発見、2種のサイトカインシグナルのクローストーク、GATAs,Evi-1などの転写因子の機能解析などの重要な発見がなされた。これらの研究成果を創薬につなげていく必要がある。第四班の「造血幹細胞の遺伝子操作」では、よりヒトに近いサルでの研究が開始されたこと、HIVベクターなどによる幹細胞への効率のよい遺伝子導入が開発された。
2)近年、神経幹細胞からの骨髄再建能をもつ血液細胞の分化することや、骨髄から骨・筋・脂肪細胞・肝臓細胞などが分化することが報告された。本研究班でも、造血幹細胞と他の系列(神経や肝臓)の幹細胞の相互変換、および骨髄にある間葉系幹細胞の多分化能という新しい領域の研究が進んだ。また、ES細胞からの血液細胞のみならず、血管内皮細胞・壁細胞の誘導をはかり、in vivoで血管を形成することに成功している。平成13年度は、この先駆的な成果を利用して、「幹細胞の多分化能」を重要課題として研究を進めたい。
