レトロウイルスベクターによる造血幹細胞への遺伝子導入とその応用

研究概要

以前、研究代表者が開発したレトロウイルスベクターpMXに加え、pMY、pMZベクターを作成し、それぞれにcDNAとしてEGFPを組み込んだコントロールベクターで、感染効率および発現を比較した。pMX-GFPはEC細胞やES細胞での発現がよくなかったが、pMY-GFPおよびpMZ-GFPはこれら未熟な細胞中でもよく発現した。
またより安全で効率のよいパッケージング細胞を作成するため、EF-Iαプロモーター-gag-pol-IRES-puroおよびEFIαプロモーター-env-IRES-bsγの構造を持つ新たなパッケージングコンストラクトを作成した。最近、これらのパッケージングコンストラクトを利用してエコトロピックパッケージング細胞を作成し、現在効率を検定中である。またアンフォトロピックパッケージング細胞も作成予定である。
現在、作成したレトロウイルスベクターおよびパッケージング細胞を利用して、恒常的活性型MPLおよびSTATの増腫瘍性をマウスの骨髄移植モデルで検討している。
