研究概要	本研究では、血球の増殖・分化・死の制御機構に関するストローマ細胞の役割を明らかにすることに加え、造血幹細胞の体外増幅の可能性を探求することを目的として、ストローマ細胞由来の造血制御因子の同定を試みた。 BMS2.4細胞株の培養上清は多くの造血細胞株の増殖を抑制するため、BMS2.4からcDNAライブラリーを作製し、WEHI3細胞株を標的としてexoression cloningを行い、増殖抑制作用を持つ新規分子の同定に成功した。この分子は182個のアミノ酸からなる未知の糖蛋白で、インターフェロンα、βとアミノ酸レベルで約20%の相同性を有していた。このcDNAを293Tに遺伝子導入して得た培養上清は、一部のpre-B細胞株にアポトーシスを導入し、他の多くの造血細胞株にG1アレストを誘導した。一方、MS-5はヒト幹細胞を長期にわたり増幅・支持しうるユニークな特徴を有するストローマ細胞株であるため、signal sequence trap methodを改良することによりMS-5由来の分泌蛋白として24種の分子を同定した。これらの中には、コラーゲン、オステオポンチン、ケラチン、プロテオグリカンなどの細胞外マトリックスやpre-PDGF受容体、トロンビン受容体などの膜蛋白の他に未知の4種類の分子や機能解析がなされていないOsteoblast stimulating factor、Placental proliferin、Hematopoieitic lineage switch-2などが同定された。現在、上記の新規分子および機能不明分子に関して機能解析を行っている。
