研究概要	ニワトリpro-B細胞株DT40から遺伝子相同組換えによる遺伝子ノックアウト法を用いてチロシンキナーゼSyk欠損変異株およびチロシンホスファターゼCD45,SHP-1欠損変異株を樹立し、その下流のシグナル伝達因子について検討した。骨髄微小環境のモデルとして各種細胞外マトリックス蛋白質をコートしたプレートを用いると、欠損変異株と野生株において接着性に有意な差が認められた。そこで、細胞外マトリックス蛋白質からの接着分子を介したBリンパ球分化制御機構を解析し、チロシンキナーゼSykおよびチロシンホスファターゼCD45,SHP-1がいかに関与しているか、これらの欠損変異株を用いて検討している。また、造血細胞分化過程における接着分子を介するシグナル伝達機構を解析するため、巨核球分化と共に発現するインテグリンαIIbβ3下流のシグナル伝達を、細胞接着とチロシンキナーゼに着目しつつ検討した。巨核芽球系細胞株CMKはフォボールエステル(PMA)処理により細胞接着と共にαIIbβ3を発現するが、可溶性リガンドとの結合は細胞接着に依存していた。また、可溶性、固相化リガンド各々により誘導されるアウトサイド-インシグナリングは、Syk、FAKなどのチロシンリン酸化において異なったが、細胞接着を介して協調的に機能していた。造血細胞分化過程において、接着分子を介するシグナル伝達と血漿中の液性因子を介したシグナル伝達が、協調的に機能しチロシンリン酸化とその制御機構が重要な役割を担っていると示唆された。
