研究概要	本計画研究班の目的は、メリステムからの植物器官・組織の発生分化の分子的背景を様々な角度から研究することである。町田は、葉の扁平性と左右対称性について、シロイヌナズナの変異体であるale1とas2を用いて研究した。その結果、前者では表皮細胞の分化が、後者では維管束の分化が異常になっていることがわかった。ale1については遺伝子クローニングを行い、その塩基配列からサチライシンファミリー,に属するセリンプロテアーゼをコードしている可能性が示唆された。AS2遺伝子については、独自の方法により染色体歩行を行い、約25kbの中の6つのORFにまで絞り込んだ。河内は、栄養成長から生殖成長への転換の分子機構を知るために、低発現遺伝子のライブラリーから花序分裂組織特異的遺伝子のcDNAを選別した。それらのうちZinc-fingerタンパク質遺伝子のT-DNAによる破壊株などを単離・解析した結果、花の異常を見いだした。島本は、短日植物であるイネの光周性を研究し、光周性を完全になくした変異体se-5の原因遺伝子がHY1である可能性を示した。塚谷は、葉の長軸方向と横軸方向の成長の制御機構を研究してきた。クローニングしたROT遺伝子の発現系を用いて、これが葉の長軸方向への伸長に関わっていることを示した。中村は、しょ糖に対する植物の応答と器官分化の関連を研究するために、7500系統のアクテイベイションタギングライブラリーを作製し、その中に、糖に対する応答が異常になった変異体を見い出した。また、しよ糖誘導性遺伝子の最小プロモーターに存在する配列を調べ、糖に応答する結合タンパク質と複数のシスエレメント(10bp,SP8a,SP8b)を検出した。西谷は、タバコ培養細胞BY-2にEXGT cDNAアンティセンスを導入し、増殖速度の遅延と細胞形態の異常を見つけた。西村は、液胞へのタンパク質輸送系を研究して、これに関与している小胞(PAC)に存在するPV100がプロセシングされ複数の機能分子になることを示した。また、栄養器官型VPEが活性を持った分子として液胞で機能していることがわかった。
