細胞質分裂シグナル伝達と収縮環形成のメカニズム

研究概要	1.分裂酵母ミオシンIIは分裂位置付近にアクチンに非依存的にドット状に集合し、これらがアクチン繊維とともに収縮環に再編成されることが分かった。次いでドット状集合に必要な部位を限定した。ミオシンV(Myo4)は分裂時に分裂溝の収縮環に局在しまた分裂溝近辺で活発に動き回る。myo4遺伝子破壊株は細胞内に顆粒を蓄積することから、分裂溝への物質輸送を行っているらしい。トロポミオシンは収縮環形成時にアクチン脱重合タンパク質Adf1と拮抗的に働くことも分かった。 2.ウニ卵にcalyculin Aで疑似分裂溝が誘導されるが、この過程の早い時期にミオシン軽鎖の活性化部位が2重リン酸化されることを示した。この結果ミオシンは表層に集積し、アクチン繊維と相互作用して表層に張力を発生させることも分かった。 3.分裂酵母の低分子量Gタンパク質Rho3が細胞質分裂に関与していることを示した。またRho3の下流に新規のdiaphanous/formin For3があり細胞の極性成長に関与することを示した。さらに新たにみいだしたRhoGAPであるRga7がRho3のGAPであることを示した。 4.細胞性粘菌の分裂必須IQGAP様タンパク質は分裂溝に集積するが、その過程はミオシンIIにもアクチン結合タンパク質cortexillin Iにも非依存的だった。さらにREMI法により新規の細胞質分裂必須の巨大タンパク質(約3,000アミノ酸)を同定した。このタンパク質はロイシンジッパーを一つ持つ以外に既知のモチ-フや相同領域を持たない。
