グリア細胞における神経伝達物質受容体の発現調節

研究概要

平成10年度および11年度の研究により、小脳プルキンエ細胞の成長樹状突起がバーグマングリアの放射状突起に接しながら軟膜方向へと伸展していく事実を見い出した(Yamada et al.,2000)。この事実は、グリア細胞の放射状突起が、1)接着性(認識性)の足場、もしくは、2)樹状突起成長を促進する栄養因子を提供することにより、ニューロン受容野の形成に関与していることを示唆している。そこで、本年度の研究では、グリア細胞によるニューロン樹状突起の誘導・形成の細胞機構および分子機構の解明と、より具体的な目標を設定した。
その結果、アミノ酸の1種であるセリンがプルキンエ細胞の生存と樹状突起伸展を促進する作用を見い出した(Furuya et al.,2000)。セリンは、体内で生合成可能であることから非必須アミノ酸に分類されている。そこで、セリン合成酵素である3-phosphoglycerate dehydrogenase(3PGDH)の発現の有無を小脳において追求したところ、バーグマングリアに豊富に存在するがプルキンエ細胞では発現を欠失していた。この事実は、プルキンエ細胞自身はセリンを合成することができず、その供給を周囲のグリア細胞に依存していることを強く示唆する。すなわち、発達段階に見られる樹状突起とグリア突起は、構造的にも機能的にも親密な関係にあることを物語っている。
