クラスリン・アダプター様タンパク質の細胞内タンパク質輸送における役割

研究概要	クラスリン被覆小胞は、トランスゴルジネットワーク(TGN)からリソソームへのタンパク質輸送や、細胞膜からの受容体分子のエンドサイトーシスなどに関与する。TGN膜や細胞膜へのクラスリンの結合は、アダプタータンパク質複合体により仲介される。TGNでのクラスリン被覆小胞の形成にはAP-1複合体が、細胞膜ではAP-2複合体が関与する。これらは,2つのlarge鎖、1つのmedium鎖,1つのsmall鎖で構成される。また最近,TGNやエンドソームで機能する新たなアダプター様複合体AP-3やAP-4も,申請者らや他の研究グループにより同定された。しかし、TGNと細胞膜との間の小胞輸送は複雑で、これら以外にも別のアダプターが存在する可能性が考えられる。本研究では新奇のアダプター様タンパク質に関して以下のことを明らかにした。 1.AP-1複合体のlarge鎖のひとつγ-アダプチンのC末端に存在するイアー・ドメインと相同性を有する3種類の新奇のタンパク質(GGA1-GGA3)を同定した。 2.GGAはいずれも,N末端側からVHSドメイン,GGAHドメイン,プロリン・リッチ領域,γ-アダプチンのイアー・ドメインと相同なドメイン(AGEHドメイン)を有していた。 3.GGAはいずれもTGNに局在し,クラスリン/AP-1コート小胞の出芽を調節する可能性を示した。 4.GGAHドメインには低分子量GTP結合タンパク質ARFが,AGEHドメインにはラバプチン5やg-シナージンが結合することを明らかにした。
