クラスリン・アダプター様タンパク質の細胞内タンパク質輸送における役割

研究概要

クラスリン被覆小胞は,トランスゴルジネットワーク(TGN)からリソソームへのタンパク質輸送や,細胞膜からの受容体分子のエンドサイトーシスなどに関与する。TGN膜や細胞膜へのクラスリンの結合は,アダプタータンパク質複合体により仲介される。TGNでのクラスリン被覆小胞の形成にはAP-1複合体が、細胞膜ではAP-2複合体が関与する。これらは,2つのlarge鎖、1つのmedium鎖,1つのsmall鎖で構成される。また最近,TGNやエンドソームで機能する新たなアダプター様複合体AP-3やAP-4も,我々や他の研究グループにより同定された。また我々は,AP-1複合体のlarge鎖のひとつγ-アダプチンのC末端に存在するイアー・ドメインと相同性を有する3種類の新奇のタンパク質(GGA1-GGA3)を同定し,TGNで機能することを示した。本研究ではこれらのアダプタータンパク質に関して以下のことを明らかにした。
1.AP-1複合体のlarge鎖のひとつγ-アダプチンやGGAが,C末端のイアー・ドメインを介して,TGNで機能するタンパク質γ-シナージンや小胞の融合に働くラバプチン5と結合することを示した。さらには,γ-アダプチンのイアー・ドメインのX線結晶構造解析を行い,γ-シナージンやラバプチン5との結合様式を明らかにした。
2.GGAのN末端にあるVHSドメインが,TGNから輸送される積み荷タンパク質であるマンノース6リン酸受容体やソーティリンの酸性アミノ酸クラスター・ジロイシン(ACLL)モチーフに結合することを明らかにした。さらには,GGAのVHSドメインとACLLペプチドの複合体のX線結晶構造解析を行い,その結合様式の詳細を明らかにした。
3.AP複合体のサブユニット間の相互作用の機構を,酵母three-hybridシステムを用いて解明した。
