神経シナプス小胞輸送の分子機構-記憶形成の理解に向けて

研究概要

神経シナプス小胞輸送の制御において、私共が発見したRab3A系とDoc2系が重要な働きをしていることが明らかになりつつある。本年度の本研究では、Rab3A系およびDoc2系の各々の作用機構について解析すると共に、Doc2系によるLong-term potentiation(LTP)の形成機構につき解析を行い、以下の研究成果を得た。
1. Rab3Aの活性制御蛋白質Rab3 GAPが、すでにクローニングしていたp130と今回新たに単離したp150の2個のサブユニットから成ることを明らかにした。
2. Rab3GAPと今ひとつのRab3Aの活性制御蛋白質Rab3 GEPが、Rab3Aと共に神経終末に局在し、Rab3Aの活性を制御することによって、Ca^<2+>依存性分泌を制御していることを明らかにした。さらに、ヤリイカの巨大シナプスの系により、Rab3Aの標的蛋白質Rabphilin-3Aが実際に神経伝達物質の放出を制御していることを明らかにした。
3. ラット上頚交感神経節細胞を用いた実験系により、Doc2αはMunc13と結合してシナプス小胞のプレシナプス膜へのdockingの過程を制御することにより神経伝達物質の放出に機能していることを確定した。
4. Doc2αのノックアウトマウスの解析により、Doc2αが海馬のシャッファ一側技のシナプスにおけるshort term plasticityとLTPの形成に機能していることを明らかにした。
5. Doc2βが微小管モーターのひとつであるダイニンの軽鎖tctex-1に結合し、微小管依存性の小胞輸送に関与していることを示した。
このように、本年度の研究は予想以上に進展し、当初の目的はほぼ達成できた。
