小胞輸送とゴルジ装置形成の分子機構

研究概要	ゴルジ局在タンパク質であるgiantin, golgin-84およびsyntaxin-5は共通してC末端に膜アンカー領域を持つ。各種欠失変異体の調製・発現実験および蛍光抗体法による解析の結果、これらのタンパク質のゴルジ装置局在化にはアンカー領域に隣接する細胞質側のcoiled-coil領域約100残基のアミノ酸配列が必須であることを明らかにした。 Giantinと相互作用を持つタンパク質の検索を行なう目的で、giantinのゴルジ体局在化に必須なドメインをbaitにして酵母two-hybrid法によってヒト細胞cDNAライブラリーをスクリーニングした。その結果、分子量60kDaの新規タンパク質を同定し、GCP60と命名した。GCP60は本来可溶性タンパク質で細胞質に存在するが、giantinとC末端領域で結合することによってゴルジ装置に局在化することがわかった。データベース検索の結果、本タンパク質は線虫(C. elegans)やショウジョウバエ(D. melanogaster)まではよく保存されているが、酵母(S. cerevisiae)にはホモログが存在しなかった。Giantinとの結合に必要なC末端領域だけを持つGCP60変異体を過剰発現すると、小胞体からゴルジ装置へのタンパク質輸送が完全に阻害され、最終的にゴルジ装置が解体された。この結果は、GCP60が小胞輸送およびゴルジ装置の構造形成に重要な役割を果たしていることを示唆する。 GCP170は我々が同定した表在性ゴルジ膜タンパク質である。各種実験によって解析した結果、GCP170はゴルジ局在化シグナルをN末端側(アミノ酸137-237)とC末端側(1120-1200)の2ヶ所に持つことがわかった。N末端側ドメインをbaitにして酵母two-hybrid法でスクリーニングを行なった結果、GCP170と結合する新規タンパク質(分子量16kDa)を同定し、GCP16と命名した。GCP16も、GCP60の場合と同じように、線虫やショウジョウバエまでは保存されているが、酵母にはホモログを見い出しえなかった。この知見は、これらのタンパク質がゴルジ装置における動物細胞特有の構造形成と機能発現に関与している可能性を示唆する。
