研究概要

1)エンドソームとオートファゴソーム:SKD1がエンドソームから細胞膜やリソソームへの輸送を制御することを示してきたが、さらにオートファジーとも関係することが判明した。エンドソームからオートファゴソームに機能素子を輸送しているものと思われる。またBeclinがphosphatidylinositol 3-kinaseと結合してTGNに局在することを明らかにした。この蛋白質複合体はTGN・エンドソーム輸送とオートファゴソーム形成に必要な機能素子供給の両方を制御すると考えている。さらにApg5-Apg12複合体がオートファゴソーム前駆体に結合し、それをオートファゴソームへ成長させていることを発見した。またApg5がApg12と結合すると、LC3-Iが翻訳後修飾によりLC3-IIとなりオートファゴソーム膜に局在化することも見いだした。以上、エンドソームとオートファゴソームに関わる機能素子を同定しその機能に関する知見が得られた。
2)血小板放出反応:Streptolysin-Oにより形質膜を透過型にした血小板で、濃染顆粒に含まれるセロトニンとα顆粒に含まれるvon Willebrand因子の放出を測定する再構成系を確立した。それらの放出反応はATP・細胞質依存的であり、細胞質中に必須因子のひとつを同定し、解析中である。また、α顆粒の放出にRab4が必須な制御因子であることを明らかにした。さらに、透過型血小板の凝集アッセイを確立し、Rhoが重要な働きをすることを明らかにした。
