研究概要	本特定領域研究は、DNAのエンドジョイニングやトランスポゾンの転移機構など、いわゆる非相同組換えの機構を「DNAの切断と再結合」という問題に絞って解析し、総合的に理解しようとするものである。本年度は、11月に3Rシンポジウム、12月の分子生物学会ワークショップ「トランスポゾン研究の新展開」及びトランスポゾンに関する研究会において関連分野の外国人研究者を招聘し、国際的シンポジウムを主催あるいは共催し、学術交流を行った。また、12月には班会議を開催し、各計画研究間の連携と調整を諮った。各班員の研究は進展しており、この領域の研究の発展に寄与した。池田班員は、大腸菌を飢餓状態にしたときに自然にDNAの二本鎖切断が蓄積し、同時に非相同組換えが亢進することを見出し、適応変異(adaptive mutation)との関連を示唆した。近藤班員は、BLMとSUMO-1が会合することを見出し、この会合が、BLMが細胞核内にドット状に局在するために必要であることを明らかにした。立花班員は、BLM及びWRN欠損細胞における染色体不安定性が相同組換えとエンドジョイニングの両方の異常によることを示唆した。松浦班員は、DNAエンドジョイニングに重要であるNbs1が胚の初期発生や細胞増殖に必須であることを示した。トランスポゾングループの研究も進展している。飯田班員は、アサガオの三種類のトランスポゾンが自然突然変異原として重要であることを示し、古賀班員はメダカのTol2トランスポゾンを遺伝子導入ベクターとして利用する系を作った。大坪班員は、大腸菌のIS3トランスポゾンの転移に働くトランスポゼースがユニークなDNA切断反応(transcatalysis)を起こすことを示した。大矢班員は、VDE遺伝子のホーミング、駒野班員は、R64シャフロン組換え系の生物学的意義を明らかにした。
