DNAの切断と再結合におけるhelicase遺伝子familyの機能解析

研究概要	本年度はBLMタンパクの機能について以下の点を明らかにした。 1)BLM欠損細胞では細胞死の亢進が認めるれる。 Bloom症候群では小柄な体型、免疫不全が認められるがその機序についての解析をおこなった。BLM-/-knock outマウスは胎生13.5日までしか生存できない胎生致死マウスであるが、このマウスの胎生12.5日の身長、体重、肝細胞の細胞数についてしらべたところ、正常マウスと比較して身長、体重が1/2程度、肝細胞数は1/100以下に低下していた。神経管周囲の細胞はsingle strand DNAの染色にて陽性であり、細胞死の亢進が考えられた。細胞増殖の低下、DNA障害因子に対する感受性の亢進により細胞死が誘導されやすく、個体の細胞数の低下が小柄な体型の原因と考えられた。 2)BLMはATM kinaseによりリン酸化をうける。 yeast two-hybrid systemを用いてBLMと会合するタンパクの同定をこころみたところ、2つのクローンが得られ一つはataxia-telangiectasiaの病因遺伝子産物ATMであった。in vitroのtranscription/translationを用いて発現させたATMまたはBLMタンパクとGSTとの融合タンパクのbinding assayではBLMのhelicase domainとATMのkinase domainが会合することが明らかになった。BLMの欠損細胞株ではradiation照射によるATMkinaseの活性が照射後1時間の時点で低下がみられ、2時間では正常レベルであった。ATMを免疫沈降しBLMを基質としてリン酸化について検討したところ、BLMのN-末の領域に存在するATMのリン酸化部位のconsensus領域のThrがリン酸化をうけることがあきらかになった。
