研究概要	Bloom症候群の病因遺伝子BLMはDNA helicase familyの一つであり、sister chromatid exchangeに抑制的に働き、homologus recombinationに関与すると考えられている。 DNAの複製・修復にはたらくタンパクの多くは他のタンパクとcomplexを形成して機能する。そこで、BLMと相互作用するタンパクをyeast two-hybrid systemを用いて検索したところATM(ataxia-telangiectasia mutated)が同定された。今までにATMと相互作用するタンパクとしてp53,BRACA1,NBS1等が同定されており、BLMと相互作用するタンパクとしてTopo-III等がしられているが、申請者が明らかにしたATMとBLMの相互作用は全く新しい相互作用である。ともに免疫不全、高発癌を示す二つの疾患が病因遺伝子産物の機能的な相互作用を通じて、共通した表現型を形成している可能性を示しており、病態解明に新たな視点をあたえるものである。 BLM-ATMの相互作用が機能的ならば、それぞれの欠損細胞株で機能に影響がみられると予想される。そこで、DT40のATM-/-のノックアウト細胞株から、ATM-/-、BLM-/-のダブルノックアウト細胞株を作製した。単独のノックアウト細胞株と細胞増殖、sister chromatid exchange(SCE)の頻度を比較したところダブルノックアウト細胞株ではBLM-/-でみられるのとほぼ同程度に細胞増殖がおそくなっていた。SCEの頻度はBLM-/-でみられるのとほぼ同程度に増加していた。ATM-/-細胞では放射線感受性が亢進していたが、ダブルノックアウト細胞株でもほぼ同程度亢進していた。以上よりATMとBLMはDNAの修復に独立の経路で機能していると考えられた。
