研究概要

江戸時代に自然突然変異体として分離されたアサガオの変異の大部分は、我々が既に単離したEn/Spm系のTpn1類縁の非自律性転移調節因子の挿入による変異であり、易変性の変異形質を示さずに安定な変異であると考えられる変異の中にも、対応する自律性因子がジーンサイレンシングによりその活性が抑えられ、その結果挿入されたTpn1類縁の非自律性因子が転移脱離できなくなり、一見安定な変異の様にふるまう変異もあることを明らかにしてきた。現存するアサガオの多様な変異体は全て自然突然変異体であると考えられるので、そのほとんどがTpn1類縁の挿入変異体と推定できる。このTpn1類縁因子の挿入によりタグされた未知有用遺伝子を簡単に同定できる簡易トランスポゾンディスプレイ(Simplified Transposon Display)法を我々は開発し、紫地に青色の絞り花を咲かせる易変性「紫」変異purple-mutableに応用した。その結果、この易変性「紫」変異は、花弁表層の液胞の膜タンパクで、液胞型Na+/H+交換輸送体をコードしている遺伝子内に、Tpn4と名付けたTpn1類縁因子が挿入した変異であることを明らかにできた。アサガオゲノム中には約800コピーのTpn1類縁因子が散在しているにもかかわらず、易変性「紫」変異の同定に成功したことから、本法が極く最近相次いで発表された類似の諸手法と比較して、簡便かつ有効的な方法であると思われる。また、花色発現に花弁液胞内のpHが重要なことは従来から論議されてはいたが、花弁液胞のpHを上昇させて花色を青色化させるタンパクの実体は、本研究によりその一端が初めて明らかにされた。
