研究概要	CFTR(Cystic Fibrosis Transmembrane Conductance Regulator)は、ABC(=ATP binding cassette)蛋白の一員であり、欧米に多い遺伝病である嚢胞性繊維症で機能異常を来すクロライド・チャネルをコードすることが、1990年前後から明らかとされた。われわれは、このCFTR isoformが心筋細胞にも発現しており、primary cultureの心筋細胞はパッチクランプ法による電流解析に適しているため、格好の研究材料を提供することを報告してきた。今回我々は、この実験系を用いてチロキシンキナーゼ(PTK)阻害薬のゲニスタインによるCFTRチャネル蛋白の活性化の機序を検討した(論文:Obayashi et al,1999)。Micromoleレベルのゲニスタインは、濃度依存的にCFTR電流を活性化したが、同時に、ゲニスタインはイソプロテレノール(ISO)によるCFTRの活性化を促進した。同様の促進作用は、PTK阻害作用のないゲニスタインのアナログであるダイドザインでも観察された。また、ゲニスタインの作用はプロテインキナーゼA(PKA)の阻害剤であるH-89存在下では、消失した。フォスフォチロシン・フォスファターゼ阻害薬であるvanadateを細胞内投与した状態でもゲニスタインは、CFTRを活性化した。一方、ゲニスタイン以外の知られているPTK阻害薬のlavendustin A,tryphostin B42,51には、CFTR活性化作用は認められなかった。以上から、ゲニスタインは、PTK活性を介するのではなく、チャネル蛋白の燐酸化に関わるPKAに依存することから、直接の作用と考えられた。
