研究概要	申請者らはこれまで、1980年代より始まった心臓や血管由来の新しい心臓血管ホルモンの生体内での意義の解明を目的として、ナトリウム利尿ペプチドファミリー、エンドセリンファミリー等の生理的臨床的意義等の研究を行い、内分泌臓器としての心臓・血管の意義や、心臓血管ホルモンの特徴を明らかにしてきた。特にナトリウム利尿ペプチドに関する研究では、ANPとBNPがそれぞれ心房と心室に由来する心臓ホルモンであること、一方CNPが血管内皮細胞で産生される局所調節因子であることを明らかにした。本研究は、以上の研究成果を踏まえて、CNPに次ぐ新たな内皮由来血管弛緩因子として注目されるアドレノメデュリン(AM)の生体内での役割の解明と臨床応用を目指すことを目的とするものである。本年度は以下の研究成果を得た。 I. 血管発生・分化・リモデリングにおけるアドレノメデュリン(AM)の意義の検討本課題ではFACSを用いたce11 sortingにより新しい血管前駆細胞(Vascular Precursor Cells;VPC)を同定し、このVPCを用いin vitro、in vivoでの血管発生モデルを開発し、そのモデルを用いAMの血管発生・分化における生理的意義を検討する。今回、京都大学分子遺伝学教室西川伸一教授との共同研究により、マウスES細胞よりflkl(中胚葉マーカー)+の細胞をFACSを用いてソーティングし、この細胞群をIV型コラーゲンコートディッシュ上で再培養することで、内皮細胞(VECadherin+、PECAM+)を分化誘導することに成功した。更に、同様の方法を用いPDGF等増殖因子添加再培養により、血管平滑筋細胞マーカーであるαアクチン+の細胞の誘導にも成功した。現在、これら前駆細胞のin vitro3次元培養、及びin vivoヌードマウスへの投与を行っている。 II. 腎におけるアドレノメデュリン(AM)およびその受容体の病態生理的意義の検討本課題では、腎糸球体や尿細管に発現し、autocrine/paracrine調節因子として働く可能性が想定されるAMの生理的役割を明らかにする。今回、AM特異的モノクローナル抗体を作製し、AMがメサンギウム細胞から分泌されること、及び強力な細胞増殖抑制とともにMAPキナーゼ活性を抑制することを見出した。一方、その受容体はCGRP/AM共通受容体に修飾膜蛋白としてのRAMPファミリー(RAMP2,3)がassociateして形成されることが報告された。現在、ラット、マウスhomologのdoningをすすめており、一部doneを得た。更に、糸球体傷害・硬化モデルとしての5/6腎摘モデル、増殖性腎炎としての抗糸球体基底膜腎炎モデル、及び間質線維化としての尿管結紮水腎症モデルを確立し、これらモデル動物腎においてAM発現が変化することを確認した。
