道管形成過程での液胞崩壊の研究

研究概要

本年度は、ヒャクニチソウ葉肉細胞から管状要素への分化転換系を用いて、プログラム細胞死過程での液胞の崩壊機構、液胞内への分解酵素のターゲッティング機構、さらには液胞崩壊後の分解酵素の働きを明らかにすることを目的として研究を行い、以下の成果を得た。
1.液胞崩壊機構の解析:
核と細胞質を染色し、連続観察した結果、管状要素成熟過程で液胞が核を押しつぶすように膨張すること、すなわち液胞が膨圧を持っている可能性があること、さらに液胞の物理的崩壊に1〜2分先だって液胞膜の選択的透過性が失われることが明らかになった。これらの結果は、液胞の性質が崩壊に先立って大きく変わっていることを示した。
2.液胞への分解酵素のターゲッティング機構とその機能の解析:
これまで、液胞崩壊時に分化特異的に作用する加水分解酵素として、ZEN1ヌクレアーゼ、ZCP4システインプロテアーゼ、ZRNase1ヌクレアーゼが知られていたが、マイクロアレイにより新たにある種のリパーゼ遺伝子が管状要素分化に伴って分化特異的に発現することが分かり、リパーゼが液胞崩壊時あるいはその後の細胞内容物の分解に関与していることが示唆された。分解酵素の細胞内局在をZEN1抗体を用いた免疫電顕法により解析した。その結果、ZEN1酵素はゴルジ体、小胞、液胞中の繊維状構造に局在していることが示された。また、E64dを用いてオートリソゾーム様の構造を誘導して観察したが、そこにはZEN1酵素は含まれず、ZEN1酵素はゴルジ体から通常の輸送系に乗って液胞に運ばれると考えられた。
