道管形成過程での液胞崩壊の研究

研究概要	我々は高等植物細胞の形態形成における液胞の寄与について研究を進めている。高等植物細胞の液胞はさまざまな機能を持ち、形態についても多様であることが知られている。近年、一個の細胞内において液胞の機能が分化に伴って変化することや機能の異なる複数の液胞が共存することが分かってきた。一方、細胞の形態形成や細胞分裂のような現象に液胞がどのように関与しているかについてはまだ不明な点も多い。今年度は生細胞における液胞の動態を可視化すべく、脂溶性の蛍光色素であるFM4-64によるパルスラベルを行い、タバコ培養細胞BY-2の細胞周期各期における液胞の動態の観察に成功した。また、GFP-α-チューブリン融合タンパク質を恒常的に発現するタバコ培養細胞の形質転換細胞BY-GT16を用いて、液胞の動態を微小管構造と合わせて検討した。これらの観察の結果、G2期の終わりよりフラグモソーム中にチューブ状の液胞構造が連なったネットワークが微小管の分裂装置を取り巻くように生じること、後期以降ではチューブ状構造はフラグモプラスト近傍にも出現すること、G1期初めに細胞板と娘核の間にある液胞構造のいくつかは核が細胞板から離れて行くにつれて体積を増加して大きな間期の液胞に成長することなどが明らかになった。また、我々はこれまでにヒャクニチソウ葉肉細胞から管状要素への分化転換系を開発し、道管形成の分子機構について詳細な解析を行い、液胞が細胞の自己分解過程で崩壊することによりプログラムされた細胞死の中心的役割を担うことを明らかにしてきた。その過程で、細胞死特異的発現を示すS1ヌクレアーゼ遺伝子(ZEN1)の産物が液胞に特異的に蓄積されることも明らかになった。今年度は、このS1ヌクレアーゼが液胞崩壊後の核分解に直接関与していることを、1)in vitroの単離核分解活性のZEN1抗体による阻害、2)アンチセンスZEN1によるin vivoでの核分解阻害から明らかにした。
