研究概要	Arabidopsis液胞膜タンパク質の分子カタログの作成のために、Arabidopsisより液胞膜を含んだ画分を分取し、高度に精製した液胞膜の調製を行っている。また、二次元電気泳動についても現在方法を検討中である。セイヨウナシ果実から液胞膜を調製し、液胞膜に特異的であると思われる数個のポリペプチドを特定することが出来た。このポリペプチドのN末端アミノ酸配列から糖輸送体の候補者を捜す。セイヨウナシ果実から液胞膜を調製し、H^+-ピロホスファターゼを単離・精製することが出来た。そしてそのcDNAをクローニングし塩基配列を決めた。植物(ヤエナリ)H^+-ピロホスファターゼのcDNAを酵母で発現させる系を確立し、部位特異的変導入およびランダム突然変異誘発の2つの方法でアミノ酸置換を行い酵素機能への影響を解析した。その結果、生物種間で保存されている特定の酸性・塩基性アミノ酸が活性に必須であることを明らかにした。全体像を得るには、より体系的な変異解析と、これとは別に三次元構造の解明も課題となってきた。セイヨウナシ果実から液胞膜を調製し、H^+-V-ATPaseを単離・精製し,11個のサブユニットの存在を確認した。そのうち5つのサブユニットにつきネズミ抗体を作成することが出来た。ダイコンを材料に細胞膜型8種、液胞型2種の水チャネルcDNAの配列を決定し、植物体での器官別、生育段階別の発現レベル、水チャネルのタンパク質量を解析し、それらが器官及び生育に伴って顕著に変化することを明らかにした。
