植物生活環の中での液胞膜輸送ネットワークの機能調節

研究概要

液胞膜タンパク質のカタログ化
シロイヌナズナ緑色植物体から、密度勾配遠心法により液胞膜濃縮画分を調整した。液胞膜濃縮画分に、塩や界面活性剤を処理し膜表在性タンパク質を除去して、膜に強く結合したタンパク質を集めた。このタンパク質をSDS-PAGEで分離し、バンドを分子量ごとにプロテアーゼ消化、逆層HPLCで分離し、各ペプチドのアミノ酸配列を決定した。その結果、合計96個の遺伝子が特定され、その半数が輸送体関連分子であった。その中には、既知の液胞膜タンパク質の他に、他のオルガネラ局在が報告されているもの、また機能や局在が不明な分子が含まれた。これらタンパク質が本当に液胞膜に局在するかを検定するために、50分子に対する抗ペプチド抗体を作製中である。
液胞膜Ca^<2+>/H^+対向輸送体
植物細胞Ca^<2+>ホメオスタシスに関与する液胞膜Ca^<2+>/H^+対向輸送体cDNAを、イネから3種(OsCAX1、OsCAX2、OsCAX3)単離し、Ca^<2+>輸送活性を酵母発現系で確認した。このうちCa^<2+>選択性の高いOsCAX1についてイネの根と葉でタンパク質量を比べると、葉で顕著に多く、しかも光照射で酵素量が減少し、培地Ca^<2+>により発現が促進されることを見出した。
水チャネルの植物ホルモン応答性
ダイコンの液胞型水チャンネルの発現を、細胞膜水チャネル(PAQ1型、PAQ2型)とともに調べた。細胞膜水チャネルは、実生根の中心柱と内皮、生長塊根では篩部で発現が高く、水ストレスおよび植物ホルモンへの応答性はPAQ2型のみで見られた。液胞膜型はどちらかといえば、恒常的にどの組織でも一定レベル以上発現した。以上のように水チャネルが、構成的発現グループと、環境応答型グループに2分されることを見出した。
