研究概要

昨年度の得られた最大の成果は、オートファジーに必須な遺伝子のうちこれまで機能が不明であったAPG6,APG14の機能が明らかになったことである。APG6はAPG遺伝子群の中で唯一既知の遺伝子と相同で液胞タンパク質の選別輸送に関わるVps30と相同であった。これまでの研究からApg6/Vps30はApg14と相互作用をするが、Apg14はVps経路には何らの異常を示さないことからApg6は2つの経路に共通の、Apg14はオートファジーに特異的に結合している因子であることが示唆されていた。今回Apg6を含む複合体を免疫沈降によって精製し、特異的な成分を質量分析法によって解析した結果、Apg6,Apg14はPl3キナーゼVps34-Vps15と複合体を形成していることが明らかとなった。このことはイノシトール3リン酸(Pl3P)の形成がオートファジーに必須であることを示している。Pl3キナーゼはVps経路に働く複合体(Vps34-Vps34-Vps30-Vps38)とオートファジーに特異的な複合体(Vps34-Vps15-Apg6/Vps30-Apg14)が使い分けられている。今後Pl3がオートファゴソーム形成のどの段階でいかなる役割を果たしているかが大きな課題である。
これまでクローニングに成功していなかったApg2をその胞子形成不能を相補することを指標にクローニングすることに成功した。APG2は1592アミノ酸からなる178Kのタンパク質をコードしていた。その細胞内局在を調べた結果、液胞近傍の構造体に他の一群のApgタンパク質と共に存在しオートファゴソーム形成に作用していることが明らかとなった。
