研究概要

オートファジーによって選択的に壊されるタンパク質の探索を網羅的に行った。2次元電気泳動による多数のタンパク質の分離の条件を詳細に検討し、非常に高い再現性を持ち多数のタンパク質を分離できる系を確立した。この手法にもとづきに飢餓条件下にオートファジー不能変異株では分解を受け難いスポットを質量分析によって同定して、幾つかの有力な候補を得ることに成功した。中でも細胞質アセトアルデヒドデヒドロゲナーゼ(Ald6p)は窒素源飢餓下に特異的に液胞で分解することが明らかとなった。この酵素の分解をモデル系として詳細な解析を進め、選択的なオートファジーの分子メカニズムの進展が期待される。
高等植物におけるオートファジーの生理的意義を明らかにする目的で、酵母オートファジー必須遺伝子のオーソログ、AtAPG8,AtAPG4の発現解析を行なった。全9種のAtAPG8および全2種のAtAPG4はシロイヌナズナのほとんどの器官で発現しており、窒素飢餓条件下で発現が誘導された。このことは全ての器官においてオートファジーが起こることを示唆している。また、GFP-AtAPG8融合タンパク質は細胞質中のドットおよびリング状構造に局在し、窒素飢餓に伴い液胞への移行が観察された。オートファジーを簡便にかつ特異的にモニターする系を確立することが出来たことから、今後、植物のオートファジーに関してさらなる知見が得られることが期待される。
