運動ニューロンのidentityを反映する分子群探索

研究概要

我々の脳は、多種類且つ多数の神経細胞がそれぞれ軸策と樹上突起を伸ばし、特異結合を形成した極めて複雑な組織であるが、発生過程を見れば、個々の神経細胞による細胞間相互作用を経た細胞識別の積み重ねにより形成される。この研究計画では、これまでに最も詳細にネットワーク形成の現象が研究されてきた実験系の一つである運動ニューロンをモデルとして取り上げ、運動ニューロンとして殆ど全ての分子的発現様式は同一と考えられるなか、それぞれのサブタイプがどのような分子群を発現することにより細胞識別を可能にし、神経支配における不連続的なidentityを表現できるのかを明らかにすることを目的とする。
細胞識別は細胞表面の膜蛋白や細胞から放出される成長因子により制御されているため、このような分子のcDNAを選択的にクローニングするシグナルシークエンストラップ法を用いることとして、今年度はその準備を完了した。変異酵母と発現ベクターをジェネンテク社から入手し、ニワトリ胚レンズcDNAから得られた100個の酵母を解析したところ、シグナル配列を持つ5個の未知分子を同定出来たことから、この方法が大変に有効な方法であることを確認した。次に、本来の目的である運動ニューロンを解析するため、ニワトリ5日目胚脊髄の胸部と腰部脊髄から、ニワトリ胚運動ニューロンに特異的に発現される細胞接着分子SC1に対するモノクローナル抗体を使用したパンニング法により、運動ニューロンを精製し、そのRNAを回収した。
