植物における体細胞多倍数性(polysomaty)の解析とその遺伝育種学的評価

研究概要	有用植物を対象として、フローサイトメトリーによるpolysomatyの存在を広範に調査し、この現象を有する植物の共通的な特性を調査した。その結果、polysomatyは多肉化した組織を持つ植物には普遍的に存在するが、多肉化しない植物でも、ナデシコ目に属する植物にはpolysomatyが共通してみられ、木本植物には少ない傾向があるなど、進化の過程で多面的な方向でpolysomatyが出現してきたことが示唆された。polysomatyは茎頂や根端の分裂組織をのぞいて、すべての器官にみられ、加齢とともにより高次の倍数性細胞が出現する傾向がみられた。人為的な倍数体を作出した場合、polysomatyのパターンは、倍数化により1段階上の倍数性に並行的にシフトするが、その際、最高次の倍数性細胞の出現が抑制されること現象が見られた。また、組織培養による植物の大量増殖過程で起こる倍数体の出現機構を明らかにするため、polysomatyとの関連について調査を行った。その結果、polysomatyを持つ組織から誘導された懸濁培養細胞は、もとの組織とほぼ同じpolysomatyのパターンを有しており、倍加した細胞も2倍性細胞と同じ頻度で分裂可能であることが示唆された。さらに培地に添加する植物成長調節物質を変えることにより、polysomatyのパターンが変化することから、植物ホルモンが染色体倍加の大きな要因となっていることを明らかにした。さらに、本研究では、DNA含量というマクロな指標が分類群の識別に有効であること、またDNA含量の異なる種間の雑種判定、さらには倍数性雑種における母親の同定など幅広い応用が可能であることを同時に明らかにした。
