絶対寄生菌の寄生性獲得戦略および非宿主抵抗性発現機構の解明

研究概要	1. 病原菌の環境適応戦略の解明 Erysiphe属Erysiphe節とMicrosphaera属との系統関係を明らかにするため、rDNAITS領域の塩基配列データを用いて系統樹を作成した結果、両属の近縁性が支持された。しかし、Erysiphe属菌とMicrosphaera属菌はそれぞれ単系統のクラスターを形成せず、互いに混ざり合った複数の小クラスターを形成した。このことは両属の分化が1回の出来事によって起こったのではなく、2回以上にわたって独立に起こったことを示唆した。 2. 病原性獲得機構の解明 (1) 病原菌分泌物の細胞生物学的、分子生物学的解析ムギ類うどんこ病菌分生胞子が疎水性膜に接触すると、直ちに液状物質を分泌することを顕微鏡観察で実証し、この液状物質は細胞壁分解酵素セルラーゼ、キシラナーゼ、ペクチナーゼを含むことを明らかにし、これらの酵素に接触した宿主の生理的変化を検討中である。比較のために、トウモロコシ炭そ病菌分生胞子のたんぱく、糖質を含む分泌物も解析し、形態形成との関連性を検討した。 (2) 病原菌の受容性誘導因子の解明ムギ類うどんこ病菌を接種したオオムギ子葉鞘から抽出した低分子水溶性画分中に、非病原菌の感染を誘導するニンヒドリン陽性物資を見い出した。病原菌が宿主に定着すると、受容性誘導因子が宿主内で合成されることを示唆した。 3. 非宿主抵抗性発現機構の解明糸状菌に対する植物の侵入抵抗性には細胞骨格の一つであるアクチン繊維が関与することが明らかになった。そこでアクチン遺伝子をクローニングし、パーティクルガンを用いてオオムギ子葉鞘細胞で過剰発現させたが、抵抗性の増強は認められなかった。一方、細胞内に大量に存在するとアクチン繊維を破壊するプロフィリンを過剰発現させると抵抗性が抑制された。以上の結果から、抵抗性発現にはアクチンが重要であることはほぼ間違い無いが、アクチン繊維の構築状態の調節など高次の制御が最も重要であることが示唆された。
