絶対寄生菌の寄生性獲得戦略および非宿主抵抗性発現機構の解明

研究概要

1)疎水性基質に接触したムギ類うどんこ病菌分生子の直下に出現する液状物質(Extracellular Matirix:ECM)の意義を解析した。胞子はECMによって疎水性基質に付着し、ECM出現領域から発芽すことによって発芽管を基質表面に確実に伸長させることを明らかにした。また、ECM中のエステラーゼが宿主表面の物質を分解し、胞子への分解産物の取り込みが発芽を促進させる要因となることを示唆した。宿主への胞子付着は感染の第一歩であり、ECM出現が本菌の感染効率を高める手段になっていることを示した。
2)病害ストレス応答におけるRacの機能を解析するため、NtRac3を過剰発現する形質転換タバコを作出した。この結果、形質転換体では、感染特異的に誘導されるPR-1aが自発的に発現誘導されることが明らかになった。また、これらの形質転換体にはアクチン細胞骨格の構築状態変化や活性酸素の生成は認められなかったが、サリチル酸の蓄積およびサリチル酸に対する感受性の増高が認められた。以上の結果より、Racはサリチル酸経路を介した防御応答のシグナル伝達に主に関与することが示唆された。
3)18S rDNAの塩基配列およびD1、D2領域を含む28S rDNAの部分配列を用い、子嚢菌類内におけるウドンコカビ目(Erysiphales)の系統関係を推察した。ウドンコカビ目は他の子嚢菌類に対して明瞭なクレードを形成し、単一の祖先種から派生したことが示唆された。ホネタケ目のMyxotrichaceae科はウドンコカビ目とともに一つのクレードを形成した。このErysiphales/MyxotrichaceaeクレードはLeotiales、Cyttariales,Thelebolaceaeなど、一部の盤菌類とも近縁であった。分子時計を用いた計算によれば、ウドンコカビ目とMyxotrichaceaeは1億9千〜1億3千万年前に分かれた。また、ウドンコカビ目内での最初の分岐時期(1億4千〜9千万年前)は被子植物の大規模な放散と一致した。
