研究概要

1.ウイルス感染の初期段階の侵入過程において重要なウイルス粒子膜と細胞膜との融合の強弱とウイルスの病原性との関連を明らかにするために、感染細胞の細胞融合を惹起するpHの範囲を強毒株(Hantaan 76-118 clone1)と弱毒株(Hantaan 76118 mu11E10)とで比較した。両株いずれもpH6.5以下で細胞融合を発現したことから、病原性と膜融合発現活性の強弱とは関連していないと考えられた。強毒株感染細胞では弱毒株感染細胞に比べ胞融合発現が僅かに早く出現する傾向が観察されたが、蛍光抗体法による染色像から、強毒株感染細胞ではより多くの抗原が発現することが原因と考えられた。
2.強毒株(Hantaan 76-118 clone1)感染SCIDマウスに、感染3週後のBALB/cマウス由来脾臓細胞を移入したところ、感染3週目までに脾細胞を移入した場合は耐過するが4週以降に移入した場合全例死亡した。しかし、感染3週目に移入した場合、移入4日目から著しい体重減少が一過性に認められ、免疫関連の病原性発現機構が確認された。
3.Hantaanウイルスの核蛋白遺伝子(S分節)とエンベロープ蛋白遺伝子(M分節)をクローニングし、哺乳動物細胞(293T細胞)で高率に発現させることに成功した。
4.Hantaanウイルスの ポリメラーゼ遺伝子(L分節)の発現のために全長のクローニングが行われた。
5.Reverse genetics法の本ウイルスでの確立のために、今後、ポリメラーゼ遺伝子クローニングの確認と発現の確認が必要である。
