研究概要	ある種のウイルスはCTLの攻撃に対して、抵抗力を高めるような蛋白を感染細胞の中に産生させるものがある。例えば、牛痘ウイルスにはCrmA遺伝子が存在し、その蛋白を感染細胞で発現する。CrmA蛋白はCTLから注入されたgranzyme Bの活性化を抑制するとともに、Fas抗原からのシグナル伝達でcaspase 1、8を阻止し、アラキドン酸カスケードでセラミドの活性化も阻害する。その結果、感染細胞がアポトーシスに陥ることを阻止する。したがって、CrmA遺伝子を移植臓器に導入することにより、拒絶反応に対して抵抗性が高まるものと考えられた。ラットの同種肝移植モデルにおいて、ドナー肝にCrmA遺伝子を導入し、拒絶反応に対する防御効果を検討した。CrmAを発現させるベクターとして、Cre酵素により制御を受けるカセットを有し、自己複製能(E1/E3領域)を欠損させたアデノウイルス・ベクターAxCALNLCrmAを用いた。すなわち、AxCALNLCrmAとCreリコンビナーゼを発現するアデノウイルスAxCANCreを共感染することにより、AxCALNLCrmAの中でcrmA発現を抑制しているスタッファー遺伝子がはずれて、CrmA蛋白が発現するようになる。この方法では、AxCANCreの投与量を調節することにより、CrmAの発現量を調整することができる。AxCALNLCrmAとAxCANCreをそれぞれ109pfu静脈内投与すると、肝で80%の肝細胞がCrmA蛋白を発現するようになる。その肝をレシピエントに移植したところ著明な生着延長効果が得られ、拒絶反応に対する予防効果が明らかにされた。
