研究概要	線維芽細胞と細胞外基質との接着性には基質や細胞との特異性に基づく差異が存在し、歯肉線維芽細胞と免疫担当細胞との接着にはVLAインテグリン、CD44/ヒアルロン酸、LFA-1/ICAM-1以外にも接着制御のシグナル伝達を担うCD43や約45 kDaの糖タンパクが関与した。TGF-βは増殖歯肉由来の線維芽細胞に多く、その線維芽細胞はLPS刺激によって多量のIL-6を分泌した。LPS投与ラット歯周炎モデルでは、炎症細胞のみならず接合上皮から白血球走化性因子が産生された。歯肉溝滲出液中のhepatocyte growth factorの量とprobing depth、gingival indexおよびIL-1β量との間に有意な相関があった。歯肉線維芽細胞に発現するPGE2レセプターのうち、EP2とEP4はICAM-1発現の抑制に関与した。重度歯周炎患者末梢血単核球をヒトHSP60で刺激するとIFN-γの発現がmRNAおよびタンパクレベルともに有意に高くなり、P.gingivalis(Pg)GroEL刺激でIL-4産生は抑制される傾向があり、歯周炎局所にHSP60、Pg GroELを認識するT細胞が集簇していた。歯周炎歯肉中の単核球におけるINF-γ量の割合は末梢血よりも低く、IL-4量のそれは高く、また、CD28、44、54陽性細胞の割合は数%であったが、CD58は圧倒的に多かった。歯周炎歯肉中のリンパ球のアポトーシスは、シグナル伝達の悪さやアポトーシス抑制分子の働きによって回避されていた。A.actinomycetemcomitans(Aa)による免疫モデルにおける腹腔再感染では局所でTNF-α、IL-6は増加し、IFN-γは大幅に増加した。Pg、Aa、Fnの1回投与の初期における局所でICAM-1、VCAM-1、CD62p、CD62Eの発現に大差はなかったが、Pgが最も早期に発現が消失した。若年性歯周炎の原因菌として注目されているAaは、a〜eの5つの血清型に分類され、これらの特異多糖を合成する遺伝子群を全て同定し、その構造と機能を解析し、この配列に基づくPCR法による血清型の同定法を開発した。
