新奇に見出した1価陽イオンポンプ触媒鎖N端の多重リン酸化のポンプ機構における役割

研究概要

胃上皮粘膜のH^+ポンプ、細胞膜のNa^+ポンプ等はP型ポンプと称され生体内における最大のエネルギー消費系のひとつである。H^+ポンプと胃潰瘍、Na^+ポンプと本態性高血圧、血液の体積調節、脳、心臓及び腎臓の機能、Ca^<2+>ポンプと筋委縮症、Cu^<2+>とウイルソン、メンクス等の人間の疫病とポンプの関係を示す重要な情報も得られてきた。これらポンプの細胞レベルでの活性は神経伝達物質、オータコイド、ホルモン等の刺激により調節を受けることが報告されてきた。しかし、これらの刺激とポンプ活性増減の間には大きな黒箱が存在し、その間のメカニズムは全く不明であった。我々はポンプATPase分野で初めてH^+ポンプのCa^<2+>に依存したSer-27残基のリン酸化とCa^<2+>に依存しないTyr-10とTyr-7のリン酸化を発見し、さらに昨年度、in vivoでのこれらTyr-残基の可逆的リン酸化を直接証明した。さらにこれらSer及びTyr残基の可逆的リン酸化に直接関与する酵素系の同定にも成功した。すなわちSer-27のリン酸化は約86kDaのPKCαとPKCβIIによりTyr-10とTyr-7のリン酸化は約60kDaのSrc-kinase、さらに約260kDaのTyr-phosphataseと、〜35kDaのpl-IによりSer-Pの脱リン酸化の生じることが示された。これら酵素系が、H/K-ATPaseの触媒鎖付近の膜にうめこまれていることも明らかになった。一方、H^+ポンプの形成するリン酸化酵素には、高濃度ATP存在下ではEPと当モルのATPが結し、EP:EATPが定常状態のH^+-ATPaseの反応中間体に存在することが初めて明らかになった。1モルのEP分解に伴って2モルのPiの遊離が生じることなどから、EP:EATPは次のサイクルでEP:EATPを形成することが示された。
