全方位蛍光集光2光子励起レーザー顕微鏡の開発と細胞内Ca^<2+>動態と作用の解析

研究概要	1.通常の落射蛍光法に加えて、反対側より蛍光測定の為の対物レンズ微細上下動装置と明視蛍光測光切替装置とモノクロメーター分波光量測定装置を制作した。今後は2光子励起全方位蛍光測光により画像測定の実用試験に入る予定である。 2.運動神経反復刺激(10〜20Hz、数分)により、ライアノジン受容体が、数分の時間経過で活性化準備(プライミング)され、次のCa^<2+>流入により活性化され、インパルスによる細胞内Ca^<2+>上昇を数倍に、伝達物質の放出を数10倍に促進し、更なるCa^<2+>流入により不活性化されること、この促進はライアノジンやサプシガーギンにより抑制されること、短期可塑性の内、Facilitationは不変だが、AugmentationとPotentiationが顕著に増大し、その後の促進の減衰は脱プライミングによることが明らかになった。さらに、Ca^<2+>チャンネルとライアノジン受容体と開口放出機構が80nm以内の距離に機能的に連関しており、Ca^<2+>貯蔵オーガネラがシナプス小胞である可能性が示唆された。 3.ウシガエル交換神経節ニコチン性シナプスで、同様の条件刺激により、興奮性シナプス後電位とインパルスによるシナプス前終末内のCa^<2+>上昇が顕著に数時間以上促進され、サプシガーギンで抑制されるがライアノジンでは阻害されないこと、またFacilitationは不変であることが明らかになった。 4.ウシガエル交換神経節細胞で、活導電位によるCa^<2+>流入により細胞膜直下でライアノジン受容体が活性化され、Ca^<2+>遊離が起こり、BKタイプのCa^<2+>依存性K^+チャンネルを活性化し、スパイクの下降を促進し、更にSKタイプのCa^<2+>依存性K^+チャンネル活性化し、緩除後電位を発生する事が解った。
