研究概要	1.幼弱ウサギを用いたワクチン株の有効性の検定:生後1〜5日の幼弱ウサギがEHECの感染系として使用できることを確認した。この系を用いてワクチン候補株として構築した由来株VT2産生性O157:H7、及び、VT1産生性O26:H11、ブタ由来VT2vp1産生性O139:H1感染性及びワクチン株としての有効性を調べた結果、良好な成績を得た。 2.幼弱ブタを用いたワクチン株の有効性の検定:幼弱ブタを用いたワクチン候補株として構築したブタ由来VT2vp1産生性O139:H1の感染性及びワクチン株としての有効性を調べた結果、良好な成績を得た。 3.ヒト由来O157:H7由来ワクチン候補株の構築:ヒト由来のEHEC O157:7株より約1kbのVT遺伝子を含み、前後に約2kbのDNAを持つ全長約5〜6kbの目的遺伝子が得られるように制限酵素を用いて切断し、必要な断片をクローニングした。これをプラスミドベクターへ組み込み、ホスト大腸菌にトランスフェクトした。Kunkel法によりGlu^<167>GlnとArg^<170>Leuとなるようにsite-directed mutagenesisを起こさせるためのプライマーを設計・合成し、これを用いて変異を導入した。変異導入を塩基配列を決定することで確認し、この変異導入遺伝子をベクターに組み込み、ホストにトランスフォームした。トランスファーマントと親株を接合させることにより、親株の染色体上のVT遺伝子とダブルミュータントVT遺伝子間で相同組み換えを起こさせ、得られた菌株のVT遺伝子をクローニングし、塩基配列の解析により変異遺伝子に置き換わっていること、Vero細胞毒素などの生物的活性を確認し、これをワクチン候補株とした。今後ワクチン株としての有効性を調べる予定である。
