ADF/チオレドキシンによる食品低アレルゲン化法の開発と食物アレルギー治療

研究概要

本研究により、以下の研究成果を得た。
1.卵低アレルゲン化法の開発:卵の主要アレルゲンであるオボムコイド(OM)およびオボアルブミン(OVA)をチオレドキシン(TRX)で処理することにより、加熱調理や蛋白分解消化酵素に対する感受性が亢進し、未処理の抗原に比してIgE抗体との結合性が1/100以下に減少する事を示し、卵の低アレルゲン化法を明らかにした。この低アレルゲン化卵の経口投与により、卵アレルギー患者に免疫寛容を誘導して、その早期治癒を導くことが出来る可能性を示唆した。
2.卵白感作皮膚炎モデルの作成とその経口免疫寛容に関する研究:卵白アルブミン経皮感作による特異的IgE抗体産生や感作部位のアレルギー性皮膚炎惹起モデルマウスを作成し、このマウスへの卵白アルブミン経口投与が投与抗原特異的に特異抗体産生を抑制し、リンパ節のT細胞の抗原特異的IL-4,IFN-γの産生も抑制する事を示した。これらの成果は低アレルゲン化食品による経口免疫寛容誘導による食物アレルギーやアトピー性皮膚炎治療の可能性とその実験モデルを示すものであった。
3.ADF/チオレドキシンのレドックス制御作用によるアレルギー制御に関する研究:TRXはcaspase3の酵素活性維持を介してアポトーシスシグナル伝達に関与すること、Ref-1と協調して転写因子p53の活性化に関与すること、白血球の血管外漏出抑制作用を有すること、GeranylgeranylacetoneがTRXの発現と分泌を促進することを明らかにした。さらに、TRXの内因性活性阻害蛋白Thioredoxin-binding protein 2/Vitamin D3-upregulated protein-1(TBP-2/VDUP-1)を同定した。
以上の研究成果は、TRXによる食物低アレルゲン化の可能性を示し、また、TRXやその制御による食物アレルギーの新しい治療法の開発の可能性を示すものであった。
