ホルモンの分子構造と脳下垂体の構造からみた生殖腺刺激ホルモンの進化

研究概要

予定通り、坂鰓類のドチザメと、硬鱗類のシロチョウザメの二種のアルファーサブユニット前駆体cDNAの塩基配列を確定でき、アミノ酸配列の推定を行った。その結果、我々の仮説から期待された通り、ハイギョの配列は条鰭類の配列よりもはるかに板鰓類であるドチザメの配列に近かった。また、硬鱗類のシロチョウザメの配列は条鰭類よりもいくらかドチザメやハイギョに近かった。
これらの事実は、明らかに、条鰭類では進化速度が速くなったという我々の仮説を支持している。このことは同じ条鰭類でも下等なチョウザメの類のαサブユニットが、普通の条鰭類より僅かだがハイギョに近いという事実でも確かめられた。
我々の仮説は脳下垂体の構造を考えても明らかである。すなわち、条鰭類は神経性脳下垂体が樹枝状に腺性脳下垂体の中に埋没しているという、特異的な構造を持っている。これは他の魚類、あるいは四足動物の脳下垂体の構造とは大きく異なっている。むしろ、板鰓類の脳下垂体のほうが四足動物のものに構造的には近いといえよう。
魚類と呼ばれ、魚の体型を持つ動物はすべて外形的には同じように見えるが、これは生息環境によって規定されて変化し得なくなっているのであって、脳下垂体のような内部構造やホルモンの分子構造そのものは大きく異なっているといえよう。ここでは分子構造はタンパク質の一次構造を意味しているが、実はホルモンのタンパク質の立体構造は魚類から哺乳類に到るまでほとんど変化していない。
上記のように我々は動物の進化を考える場合に、どのような構造を対象としているかについて考慮することが極めて重要であろう。これが本研究で今回得られた結論である。
