研究概要	本研究最終年度にあたる今年度は、以下の3点について研究を行なった。 1)アゲハ求蜜行動の作用スペクトルを、単色光刺激への行動を分析する方法で測定した。結果、紫外線、青、赤の波長域で感度が高いこと、緑では感度が低いことがわかった。また、単色光スポットを訪問する行動のスペクトルと、スポットに対して口吻を伸ばすスペクトルに微妙な違いがあった。この事実は、訪問行動と吻伸展行動とが別々の機構によって調節されていることを示唆する。 2)アゲハ複眼の蛍光個眼について、蛍光の起源が3-OHレチノールであることは、昨年度の研究で確定した。今年度は複眼網膜内で特異的に発現する3-OHレチノール結合タンパク質を発見・同定した。アミノ酸部分配列を参考にPCRプライマーを設計、このタンパク質をコードするmRNAの全長配列を決定し、全アミノ酸配列を推定した。結果、このタンパク質はこれまでに同定されているどのレチノイド結合タンパク質とも異なる、新規のタンパク質であることが分った。 3)アゲハ個眼の基底視細胞に含まれている視物質mRNAを、in situハイブリダイゼーション法により同定した。この細胞には、複眼全域にわたって2種類の緑感受性視物質L1とL2のmRNAが重複発現していることがわかった。この研究の結果、アゲハ複眼の細胞構成はほぼ完全に解明された。 4)視葉板大単極細胞(Large Monopolar Cell,LMC)の分光感度を測定する実験系を開発した。LMC活動電位の細胞内記録と細胞内色素注入法の確立に成功、いくつかの細胞の染色像を細かく解析した。
