植物病原糸状菌における宿主特異的毒素に依存した寄生性分化の分子機構

研究概要	Alternaria alternataには、宿主特異的毒素を生産する7種の病原型が存在する。本研究では、これらのうち構造類似の毒素を生産するナシ黒斑病菌(AK毒素生産菌)、イチゴ黒斑病菌(AF毒素生産菌)およびタンゼリンbrown spot菌(ACT毒素生産菌)の毒素生合成遺伝子群の構造と機能を比較解析し、寄生性分化の分子機構の解明を目指す。本年度の成果は以下の通りである。 1.ナシ黒斑病菌のAK毒素生合成遺伝子クラスターの構造解析 AK毒素生合成遺伝子を含むコスミドクローンを単離し、約19kbの領域に6遺伝子(AKT1、AKT2、AKT3、AKT4、AKTRおよびAKTS1)が存在することを見出した。これらのうちAKTS1以外の5遺伝子が、ナシ黒斑病菌だけでなく、イチゴ黒斑病菌とタンゼリンbrown spot菌にも存在し、3毒素に共通な部分構造の生合成に関与すると推定した。 2.イチゴ黒斑病菌とタンゼリンbrown spot菌の毒素生合成遺伝子の単離イチゴ黒斑病菌から、AKT相同配列クラスターを含むコスミドクローンを単離した。単離したクローンに存在するAKT1、AKT2、AKT3およびAKTR相同配列(SAKT1、SAKT2、SAKT3およびSAKTR)の構造を解析した結果、ナシ黒斑病菌遺伝子と90%以上の塩基配列が一致し、両菌の相同配列が同じ機能を持つことが示唆された。さらに、イチゴ黒斑病菌のSAKT1およびSAKT3配列がAF毒素生産に不可欠であることを部位特異的遺伝子破壊法によって確認し、これら配列をAFT1およびAFT3と命名した。タンゼリンbrown spot菌から、PCRを用いて、AKT1、AKT2およびAKTR相同配列(TAKT1、TAKT2およびTAKTR)を単離し、それらが発現していることをRT-PCRによって確認した。
