研究概要

1.非メバロン酸経路とメバロン酸経路の両方を有する放線菌Streptomyces sp.CL190から、テルペン生合成の鍵酵素である3-hydroxy-3-methylglutaryl coenzyme A reductaseを原核生物からはじめて精製した。
2.また、本菌から1-deoxy-D-xylulose 5-phosphate synthaseを精製し、その酵素的性質を明らかにした。
3.昨年度に報告した1-deoxy-D-xylulose 5-phosphate reductoisomeraseをコードする遺伝子yaeMの破壊株を大腸菌で作成し、この遺伝子がこの酵素をコードしていることを確認した。
4.3-hydroxy-3-methylglutaryl coenzyme A reductaseをコードする遺伝子を、別のテルペン化合物生産菌からクローニングした。
5.非メバロン酸経路の生合成中間体である2-C-methyl-D-erythritol 4-phosphateから、4-(cytidine 5'-diphospho)-2-C-methyl-D-erythritolが新規酵素2-C-methyl-D-erythritol 4-phosphate cytidylyltransferaseによって生成されることを証明した。
6.上記4-(cytidine 5'-diphospho)-2-C-methyl-D-erythritolが、4-(cytidine 5'-diphospho)-2-C-methyl-D-erythritol kinaseによってその2-リン酸化体に変換されることを証明した。
7.次の非メバロン酸経路の反応段階が、上記2-リン酸化体の2-C-methyl-D-erythritol 2,4-cyclodiphosphateへの変換であることを証明した。
