高等動物の光受容細胞、生体時計局在細胞による計時機構と同調機構の解析

研究概要

地球上のあらゆる生物は地球の自転や公転が作り出す環境の周期的変化に適応すべく自己体内に計時機構(生物時計)を具備したと思われる。この生物時計は約24時間を周期とするサーカディアンリズムや約1年を周期とするサーカニュアルリズムなどを駆動し、生体内諸生理機能のリズムを制御している。本研究では、このような生体時計機構がどのようにして種々の生体機能のリズムを駆動しているのか?また、生体時計と他の本能的機能、例えば摂食機構がどのようにリンクしているかを解析した。
鳥の松果体には時計機構、光受容機構、およびメラトニン分泌機構が存在し、お互いにリンクしている。そこで、時計のリズムはメラトニンを介して生理機能のリズムを駆動している可能性が推察される。そこでメラトニンをスズメやウズラに単一投与したところ、急激な体温低下と行動抑制が認められた。このことは、これらの鳥類では夜間に松果体や網膜からメラトニンの旺盛な分泌が起こり、これにより体温低下、行動抑制、睡眠が誘起される、いわゆる外部環境リズム-生物時計リズム-生理機能リズムの流れが判明した。
我々はGH促進ホルモン、ghrelinに強力な摂食促進作用があること、さらにニューロメジンUに摂食抑制作用があることを見出した。そこで、この摂食関連ホルモンと生体時計の関係を検討した。その結果、ニューロメジンUの単一投与が、投与時刻依存性にリズムの位相変位を起こすことを見出した。さらに、時計部位である視交叉上核においてニュロメジンUの免疫陽性細胞が存在する事を見出した。以上の事実から、摂食機構とリズム機構が物質的なリンクをなしていることが推察された。
