スフィンゴ糖脂質による神経細胞の増殖・分化シグナルの制御

研究概要	酸性スフィンゴ糖脂質であるガングリオシドの神経細胞の増殖・分化の制御機構を明らかにするために、培養細胞株およびマウスにおける遺伝子操作を行い、糖鎖をリモデリングして解析を行った。ラット褐色細胞腫PC12にGD3合成酵素遺伝子を導入したトランスフェクタントにおいて観察された増殖亢進と神経成長因子(NGF)不応性のメカニズムの解析として、NGF受容体TrkAの恒常的リン酸化と二量体形成を更に検討し、全てのトランスフェクタントにおいてNGF刺激後120分以上の長時間にわたって、強いリン酸化が持続することを確認した。これらのトランスフェクタントではGD1b、GT1bの増加が見られたので、外来性に個々のガングリオシドをPC12培養液に加えたところ、ある濃度の範囲では増殖亢進に働くことが判明した。また無血清処理によるアポトーシスの誘導に対し、本トランスフェクタントは抵抗性を示したが、ワルトマニン処置で抵抗性が消失することにより、TrkAの自律的リン酸化がP13キナーゼの恒常的活性化を惹起していることが示唆された。複合型ガングリオシド、b系列ガングリオシド群によるニューロン死の抑制および切断神経の再生促進効果を検討するため、GM2/GD2合成酵素およびGD3合成酵素遺伝子のノックアウトマウスを用いて、舌下神経切断実験を行った。前者ではアポトーシス抑制および再生度において野生型の約3分の1以下に低下が見られ、後者ては共に約3分の2まで低下していた。これらの結果より、a系列ガングリオシド群の方が、舌下神経に対する神経栄養因子様活性物質としてより重要である可能性を示唆するものと思われた。現在、複合型ガングリオシド欠失マウスの脊髄後角で見られた変性のメカニズムにつき、遺伝子発現の変化を検討することにより解明中である。
