研究概要

1. ヒト胃がんと大腸がんおよび胃・大腸の前がん性病変におけるhTERT(human telomerase reverse transcriptase)の発現を、RT-PCR法を用いて検討した結果、胃がんおよび大腸がんの大部分が正常粘膜上皮より高い発現を示し、それらはテロメラーゼ活性とよく相関していた。hTERT抗体を用いてhTERT蛋白の局在をみると、hTERT蛋白は、全てのがん細胞の核に認められた。興味ある点は、hTERT陽性細胞が、がん細胞のみならず正常胃腸管下部に存在する上皮細胞および一部のリンパ球の核にも検出されたことである。
しかし、その他の間質細胞にはhTERT蛋白は存在しない。前がん性病変である胃の腸上皮化生、胃・大腸の腺腫においてもhTERT陽性上皮細胞の増生が認められた。胃の腸上皮化生、腺腫および腫瘍組織におけるテロメラーゼ活性を定量化し比較すると、化生における活性は腫瘍組織の100分の1、腺腫における活性は腫瘍組織の10分の1であった。更に、大腸の1つpit腺管を上部と下部とに分けて、テロメラーゼ活性とhTERT蛋白の発現を比較検討すると、下部の腺管のみにテロメラーゼ活性とhTERT蛋白が検出された。
2. 胃がんおよび前がん性病変(腸上皮化、過形成性ポリープ、腺腫)におけるhTR(human telomerase RNA)の発現と、in situ mRNA hybridizationを用いて検討し、併せテロメラーゼ活性との相関性をみると、hTR発現細胞は,細胞の核に存在し、正常胃粘膜、前がん性病変に比べてがん細胞に有意に頻発していた。しかし、テロメラーゼ活性hTRの発現レベルと相関していなかった。
